Use with skills which give conditions to the enemy, but make sure that the condition lasts for a very short time. This can be particularly effective against bosses with natural resistance. Virulence is perhaps the most devastating partner spell to use, especially if the linked attribute is low. Burning is often equally popular, especially from spells such as Rodgort's Invocation or Immolate. Fragility will double the damage caused by three seconds of burning (at 16 Illusion). Mark of Rodgort is a poor choice, however, as the burning often lasts for the full duration of the hex.
Fragility will only trigger on the first instance of a condition. E.g., repeatedly attacking a target while using the preparation Apply Poison will not trigger Fragility on each successive hit. (Unless the target is a dervish with Avatar of Melandru)
Crystal Wave or Teinai's Crystals can both be effective with this skill, as they do damage and "clean" the target of all conditions, triggering all of the damage at once. However, this usually requires a partner to perform effectively.
